چکیده

A current approach to the investigation of the biochemical basis for neuronal function is the development, from neuroblastoma tumors, of clonal cell lines which grow rapidly in cell culture but which retain the ability to express one or more characteristics of differentiated neurons. These characteristics may be morphological (1-3), biochemical (1, 4), or physiological (5-8). Recently, it has been shown that cells of some clonal lines derived by somatic cell hybridization between neuroblastoma cell lines and L cells are capable of stable expression of neuronal characteristics (9, 10) and that the cells of some of these hybrid lines can express certain neuronal characteristics to a greater degree than the neuroblastoma parent ceils (9, 10). The present report deals with two clonal lines derived by somatic cell hybridization between neuroblastoma and glioma cell lines ~. The cells of these hybrid lines are induced to differentiate by Ne, O 2' dibutyryl adenosine 3',5' cyclic monophosphate (dBcAMP). The differentiated cells are neuronal in light microscope appearance ~, have membranes of high electrical activity, capable of generating action poten-

A Neuroblastoma X Glioma Hybrid Cell Line with Morphine Receptors

A neuroblastoma X glioma hybrid cell line with well-developed neural properties was found that has high-affinity morphine receptors. The average cell contains approximately 3 X 105 receptors. In contrast, parent cells and other neuroblastoma or hybrid cell lines tested had few or no morphine receptors. Membrane preparations that bind narcotic analgesics such as morphine and its congeners have b...

Nuclear reprogramming of somatic cells by in vitro hybridization with ES cells

The resetting of a somatic epigenotype to a totipotential state has been demonstrated by successful animal cloning, via transplantation of somatic nuclei into enucleated oocytes. We have established an experimental system, which reproduces the nuclear reprogramming of somatic cells in vitro by fusing adult thymocytes with embryonic stem (ES) cells. Analysis of the lymphoid-cell-specific V-(D)-J...

Comparison of reprogramming ability of mouse ES and iPS cells measured by somatic cell fusion.

The ectopic expression of the key transcription factors Oct4, Sox2, c-Myc, and Klf-4 have been shown to reprogram somatic cells to a pluripotent state. In turn these induced pluripotent stem (iPS) cells, like embryonic stem (ES) cells, have been shown to be able to reprogram somatic cells by cell fusion. In this study we compare the differences and similarities between ES and iPS cells measured...

Role of human chromosome 11 in determining surface antigenic phenotype of normal and malignant cells. Somatic cell genetic analysis of eight antigens, including putative human Thy-1

The expression of eight serologically and biochemically distinct human cell surface antigens defined by monoclonal antibodies was examined on a panel of rodent-human somatic cell hybrids. Cosegregation was observed for human chromosome 11, and surface expression of all eight antigens was studied. Serological analysis of hybrids containing defined segments of human chromosome 11 permitted the re...

Synapse formation between clonal neuroblastoma X glioma hybrid

Clonal neuroblastoma X glioma hybrid cells were shown to form synapses with cultured, striated muscle cells. The properties of the synapses between hybrid and muscle cells were similar to those of the normal, neuromuscular synapse at an early stage of development. The number of synapses formed and the efficiency of transmission across synapses were found to be regulated, apparently independentl...
